1. First turn on the EVA1 spectrophotometer, so that it is heated up for later use.

. Dispense 100 µl Buffer#1 into the cuvette.

3. Followed by 2 µl sample. It is important for the outcome of the test, that the amount is precisely 2 µl. |

4. Place the cuvette in the EVA1 spectrophotometer as shown in the drawing: Arrow against arrow.

5. Let the sample and Buffer#1 heat for 10 minutes, alternatively in 37°C incubator. |

. Press the RUN (F1)...

7. Then immediately add 100 µl Buffer#2 to the cuvette.
|

8. Remove and shake the cuvette for 15 sec. with a Vortex Mixer.
|

9. Hit the cuvette gently against a hard surface to assure enough liquid is in the measuring area at the bottom. |

10. Before countdown place the cuvette in the cuvette holder of the EVA1.
|

11. Read out result after 3 minutes.
